Abstract

[ETHNOPHARMACOLOGICAL RELEVANCE] Ampelopsis japonica (Thunb.) Makino (AJ), first documented in Shen Nong Ben Cao Jing during the Eastern Han Dynasty, have been recorded for its detoxifying, heat-clearing and analgesic properties. The efficacy, application and related properties of AJ have been further supplemented and elaborated in classic works such as Ben Cao Gang Mu.

[AIM OF THE STUDY] This review aims to provide a critical overview of the traditional uses, phytochemistry and pharmacology, quality control and application prospects of AJ, with a focus on novel discovered compounds, polypharmacology, and mechanisms underlying its anti-cancer activity.

[MATERIALS AND METHODS] Literature was retrieved from PubMed, ScienceDirect, CNKI, and traditional Chinese medicine sources using keywords such as "Ampelopsis japonica", "pharmacological effects", and "chemical constituents". Notably, network pharmacology analysis was used to elucidate the multi-target regulatory mechanism underlying the anti-cancer effects of AJ.

[RESULTS] 137 compounds have been identified from AJ, including flavonoids, polyphenols, sterols, organic acids, triterpenes and so on. Studies demonstrate that AJ exhibits anti-cancer, anti-inflammatory, antibacterial, antioxidant, antiviral, and melanin-inhibition activities. Clinical studies validated AJ's efficacy in cancer therapy, burn healing and dermatology. Network pharmacology analysis revealed that flavonoids and polyphenols are the major anti-cancer ingredients and AKT1, EGFR, ESR1, BCL2, MMP9, PPARG, PTGS2 and SRC are the core anti-cancer targets.

[CONCLUSION] Modern pharmacological studies provided reliable evidence for traditional uses of AJ. Network pharmacology provides insights into its anti-cancer components and mechanisms. Further research should focus on molecular targets, quality standard establishment, and long-term toxicity to facilitate its clinical application, particularly in cancer therapy.
