Abstract

Hepatocellular carcinoma (HCC) is a common and complex malignant tumor whose etiology is influenced by a variety of factors and exhibits different outcomes during disease progression. HCC is distinguished by the triggering of various cell death pathways, for example ferroptosis, cuproptosis, and PANoptosis. This review provides insights into the current understanding of the interactions between reactive oxygen species (ROS) and these novel mechanisms of cell death in HCC, highlighting their significance in disease pathogenesis and their potentiality as therapeutic goals. We explore the biochemical mechanisms of iron and copper metabolism, highlighting the critical role of these metals in regulating ROS generation and its impact on cell viability. Furthermore, this review elucidates the impact of ROS-mediated cell death on the innovation of targeted therapeutic strategies for HCC. In conclusion, a profound understanding of the complicated relationship between ROS and cell death pathways in HCC is vital for the advancement of cutting - edge therapeutic strategies. This review underscores the potential of ferroptosis, cuproptosis, and PANoptosis as targets for ROS regulation in HCC, laying the groundwork for the advancement of precision medicine approaches to combat hepatocellular carcinoma.
