Self-assembly peptides play an important role in tumor drug delivery, promoting high drug penetration and long drug retention. With the further research on self-assembly peptides, the biophysical role of self-assembling peptides has gradually attracted attention. Self-assembly peptides form assemblies with certain configurations through noncovalent interactions. The characteristics of its assembly play a rich role in anti-tumor therapy. The physical drag effect generated by the assembly can promote the oligomerization and regulatory signals of receptors. The formation of the assembly can act as a physical barrier to inhibit material exchange and information exchange. The hydrophobicity of the assemblies might perturb the cell membrane and promote cell death. In this paper, the biophysical effects of peptide assembly are analyzed, and its intervention effect in anti-tumor therapy is further summarized. At the same time, more feasible suggestions are put forward for the design of self-assembly peptides, so that self-assembling peptides can put in a good performance in anti-tumor therapy.
