Cancer is among the most life-threatening diseases affecting the global population, and its treatment remains a major challenge in cancer management. Recently, emerging applications of immunotherapy have introduced innovative strategies for cancer treatment, highlighting the regulatory role of inflammasomes. It has been demonstrated that inflammasomes mediate cancer progression through multiple pathways such as JNK/MAPK, TLR4/MyD88/NF-κB, and organelle disruption. Besides, inflammasomes could facilitate the release of inflammatory cytokines, thereby modulating the tumor microenvironment by regulating immune cell infiltration, PD-1/PD-L1 axis, and GSDMD-induced cancer pyroptosis. Importantly, integrative approaches that combine immunotherapies and nutritional interventions have shown promising anti-cancer outcomes, where inflammasome signaling may play a key role in these processes. Finally, the dynamic roles of inflammasomes in cancer immunotherapy and tumor microenvironment are discussed in this paper. Overall, this review provides advanced updates on inflammasome-mediated cancer immunotherapeutic strategies, aiming to provide preclinical and nutritional innovations for future cancer therapy.
